TLDR; Hamas adopted the slogan in 2012. Jewish people say it is a call to genocide saying "Have no doubt that Hamas is cheering those ‘from the river to the sea’ chants, because a Palestine between the river to the sea leaves not a single inch for Israel."

It is a call to genocide, because Hamas is an openly genocidal organization, just like the current government of Israel.
